Improvements on the methodology of 19F protein label detection by NMR spectroscopy.
Abstract
19F NMR is a powerful tool that used to determine protein structure and dynamics. This tool has been applied to the study of proteins including arrestin and faced some challenges regarding label stability and experiment duration. In this work, we optimize the NMR acquisition parameters for better experiment times and limit of detection in order to use limited resources more efficiently. The stability the protein label was also investigated by time dependent NMR analysis to assess its purity and lifetime. The outcome of both of these studies is reported here.
